Reporter genes - Getting the reporter gene in cells - Stable transformation

4 important questions on Reporter genes - Getting the reporter gene in cells - Stable transformation

What is stable transformation?

Integration of the reporter gene into the genome.

What are advantages of stable transformation? (2)

1. Keep the gene in multiple generations
2. Crossing is possible --> double labelled plant lines.

How is a cell transiently transformed?

The Agrobacterium tumefaciens injects its Ti-plasmid into the plant cell after which it randomly integrates into chromosomal DNA of the plant.

How is this stable transformation done?

1. The target gene is inserted into the T-region of the Ti plasmiid and is transformed into the bacterial cell along with an adjusted Ti-plasmid only containing the vir gene (so no T-DNA).
2. The bacterium then injects the good plasmid into the plant cell.
