Polysaccharides and health - Fibers - Determination of dietary fibre

6 important questions on Polysaccharides and health - Fibers - Determination of dietary fibre

What are the two main methods to analyse diatary fibers?

  • Enzymatic/gravimetric method
  • Uppsala method

What is the main difference between the enzymatic/gravimetirc method and the uppsala method?

With the enzymatic/gravimetric method you only get the toal amoutn of fibers and with the uppsala method you can also see which fibers are present.

How does the enzymatic/gravimetric method work?

The dietary fibers are isolated, dried and weight.

A enzyme is added to remove the digestible starch.

The soluble non-starch polysccharides are removed with 80% ethanol.

How does the uppsala method work?

The sample is incubated with a thermostalle amylase and amyloglucasidase to digest starch.

Non-starch polysacharides are separated by 80% ethanol precipitation.

After drying the residue contains bout soluble and insoluble fibers.

You can split them with a filtration step.

Neutral sugars are determentin with the HPAEC.

Are all oligosacharides inclueded in the uppsala method?

No you can not relaiy on this.

Which step in the uppsala method ditermens withch fibers you have.

The preticipation step.
