  • Which contaminated Materials go into the following: White buckets, White buckets w/ orange liner, Benchtop (R2) units, Trash can

    White Buckets: Test Tubes
White buckets with liner: Petri Plates
    R2 units: Micoscope slides
Trash Can: None (Paper towels are not contaminated)
  • What color will gram - organisms be at each step

    Crystal Violet: Purple
    Iodine: Purple
    Decolorizing: Clear
    Safranin: Red
  • Why is a thick smear not ideal for staining

The organisms layer on top of each other, it is hard to see individual organisms
  • What is a CFU? Why is it used?

CFU = Colony Forming Unit. It is used because it is never certain that a colony arose from a single bacteria cell as sometimes they clump together
  • Why are agar plates incubated upside down?

So that condensation does not drip onto the plate, interfering with colony growth
  • How does a capsule contribute to pathogenicity?

    • Makes organism more hardy to environment
    • Sometimes Inhibits immune response of host cell due to composition
  • What feature does "negative staining" allow us to observe? Describe what this looks like.

Negative staining allows us to see the capsule of a bacterium. The bacteria itself is stained red, the background of the slide stains a blue color and the capsule itself retains no dye and appears as a clear negative.
  • What techniques is used for the primary staining of a spore stain procedure? Why is this used?

The primary stain was applied onto a paper towel over a steam bath. The heat is necessary in order to allow the stain to penetrate the spore coat
  • Know where the following are located on a microscope

    Eyepiece
    Nosepiece
    Objective lens
    Slide Clip
    Stage
    Condenser
    Light brightness adjustment dial
    Lamp
    Coarse focus
    fine focus
    Stage controls
  • Formula for resolving power

    Wavelength / Numerical Aperture
